Target
FUNCTION: Participates in the innate immune response to microbial agents. Detects the unmethylated cytidine-phosphate-guanosine (CpG) motifs present in bacterial DNA. Acts via MYD88 and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response.
SUBCELLULAR LOCATION: Membrane; Single-pass type II membrane protein.
TISSUE SPECIFICITY: Highly expressed in spleen, lymph node, tonsil and peripheral blood leukocytes, especially in plasmacytoid pre-dendritic cells. Levels are much lower in monocytes and CD11c+ immature dendritic cells. Also detected in lung and liver.

Storage
Maintain the lyophilised/reconstituted antibodies frozen at -20C for long term storage and refrigerated at 2-8C for a shorter term. When reconstituting, glycerol (1:1) may be added for an additional stability. Avoid freeze and thaw cycles.

Rabbit antibody to TLR9 (830-880) IF on human plasmacytoid dendritic cells. Cells were fixed with 2% formaldehyde for 10 minutes at 37°C. Washed twice with PBS before cytospin the cells onto microscope slides. Cells were blocked with PBS containing 1%BSA for 20 minutes at room temperature (RT). Excess of blocking solution was removed and cells were then incubated with Rabbit antibody to TLR9 (830-880) ( OST00297W) for 30 min at RT (diluted 1:100 in the blocking buffer). Washed 3X with PBS and incubated with anti-Rabbit Alexa 586 for further 30 minutes. Washed as before and nuclear counterstained with DAPI.

SDS-PAGE: 10%
Blocking: 1x PBS containing 3% low fat dry milk and 0.1% Tween-20 with shake for 1 hour at RT
Washing buffer: 5 times; 1xPBST (0.1% Tween-20)
Primary antibody: dilution: 1:500 incubated overnight at 4C Secondary antibody (HRP-Gt anti Rb): dilution: 1:10,000 incubated 1 h at RT
Detection: ECL on C-Digit from Licore set at high resolution
