Immunogen

Target
Function: Required for oligodendrocyte and motor neuron specification in the spinal cord, as well as for the development of somatic motor neurons in the hindbrain. Cooperates with OLIG1 to establish the pMN domain of the embryonic neural tube. Antagonist of V2 interneuron and of NKX2-2-induced V3 interneuron development.
Subcellular location: Nucleus. Cytoplasm
Tissue specificity: Expressed in the brain, in oligodendrocytes. Strongly expressed in oligodendrogliomas, while expression is weak to moderate in astrocytomas. Expression in glioblastomas highly variable.
Involvement in disease: A chromosomal aberration involving OLIG2 may be a cause of a form of T-cell acute lymphoblastic leukemia (T-ALL). Translocation t(14;21)(q11.2;q22) with TCRA.

Storage
Maintain the lyophilised/reconstituted antibodies frozen at -20░C for long term storage and refrigerated at 2-8░C for a shorter term. When reconstituting, glycerol (1:1) may be added for an additional stability. Avoid freeze and thaw cycles.

Description
Osenses Control column kit offers a convenient way to perform an On-Column-Absorption (OCA). In OCA both the bound antibody and the flow-through is utilized. The bound immunoglobulin is the antigen specific antibody and the flow-through is the negative control. OCA is an effective alternative to affinity purification and absorption at the same time.

Protocol for coupling of the control peptide to the beads

Wash>Conjugate>Use immediately or Store

Hands-on work: 15 minutes

1. Dissolve completely the peptide in 50 ┬Ál MQ water and put aside.

2. Wash the activated beads in the OS-50 spin column with MQ water. First, remove the outlet cap from the column and then place the column in a microtube.

3. Add 450 ┬Ál MQ water to the column making sure that all the beads are collected to the bottom of the tube. Discard the flow through. Repeat the washing step two more times with water. Note that the washing can also be done using the Luer cap provided and a syringe or a centrifuge (just pulse spin).

4. Cap the outlet of the spin column and add 450 ┬Ál of coupling buffer.

5. Add the peptide solution to the column and incubate for 4 h at RT (23C) or overnight at 4C while gently mixing.

6. Add 0.5 mg sodium borohydride and incubate for 30 min at RT.

7. Open the outlet cap. Wash with 450 ┬Ál of PBS pH 7. Wash two more times with 450 ┬Ál PBS.

8. The column is ready. Use immediately or add 200 ┬Ál of PBS containing 0.1% NaN3 and store at 4C.

Rabbit antibody to OLIG2 WB on mouse brain lysate (denatured, reduced) using Rabbit antibody to OLIG2 (OSO00005W) at 1: 1000 dilution; blocked with 1% LFDM for 15 minutes at room temperature with shake, primary antibody incubated for 15 minutes at room temperature, washed 3 times with PBST, 5 minutes each. Secondary antibody was also incubated for 15 minutes at room temperature.
