Target
FUNCTION: Nuclear phosphoprotein which forms a tight but non-covalently linked complex with the JUN/AP-1 transcription factor. Has a critical function in regulating the development of cells destined to form and maintain the skeleton. It is thought to have an important role in signal transduction, cell proliferation and differentiation.
SUBUNIT: Heterodimer. Interacts with DSIPI; this interaction inhibits the binding of active AP1 to its target DNA. Interacts with MAFB

Storage
Maintain the lyophilised/reconstituted antibodies frozen at -20C for long term storage and refrigerated at 2-8C for a shorter term. When reconstituting, glycerol (1:1) may be added for an additional stability. Avoid freeze and thaw cycles.

Applications
IHC, WB. Use at a dilution of 1 : 1000 to 1 : 2000. The optimal dilution should be determined by the end user. Not yet tested in other applications. For WB, the lysate must be prepared in G1 lysing buffer which is: Ripa lysing buffer containing 1% Triton X-100; 1% SDS; 1% SDC.

IHC-P on paraffin sections of mouse brain. The animal was perfused using Autoperfuser at a pressure of 130 mmHg with 300 ml 4% FA before being processed for paraffin embedding. HIER: Tris-EDTA, pH 9 for 20 min using Thermo PT Module. Blocking: 0.2% LFDM in TBST filtered thru 0.2 µm. Detection was done using Novolink HRP polymer from Leica following manufacturers instructions; DAB chromogen: Candela DAB chromogen from Osenses. Primary antibody: dilution 1: 1000, incubated 30 min at RT using Autostainer. Sections were counterstained with Harris Hematoxylin. |
